I can say for us in 2014 and in Moscow my then fiancé scheduled both her medical and interview and they were two days apart. She arrived in Moscow on Tuesday morning, had her medical exam, then picked me up from the airport as I arrived in the afternoon, we spent Wednesday sightseeing around Moscow, Thursday morning was her interview, and we did a little more sightseeing Thursday afternoon, and then departed for our then respective homes on Friday. The consulate had the results from the medical exam when she went for the interview on Thursday morning. Of course that was nearly 10 years ago, and a completely different consulate, and pre-covid so YMMV.

You should be able to update the DS260 at the interview. I am not sure what documents (i.e. custody agreements, etc.) you will need to be able to get the children visas. Good Luck! -
So you entered the US activated your LPR status and left? You can file the I407 if you wish, and be free to apply for any visa in the future you are eligible for. On the other hand, you can do nothing and gamble. Good Luck!

DENVER CO N400 Case Filers

This might be helpful (link). I have no experience with Denver, but my wife and I did have a combo interview back when she naturalized. In our case, the IO asked me to come back as well and she went over the I751 with us as well as asking if we wanted to add any additional information (we added updated tax transcripts since submitting the I751). That lasted about 5 minutes at most, then I left, and my wife and the IO proceeded with the N400 interview. Back then combo's were fairly uncommon, I am hearing more stories of folks getting their I751 approved without an interview when the N400 is also pending. That is a great point, with a marriage in October 2019 and a GC in September 2020, it definitely was not a 10yr, but rather one of the 6yr GCs. I would think ROC would tackle some of the marital living arrangement issues. That being said, as to N400, I agree with Mike E, wait for the 5 year rule.
