Does spouse need to attend marriage interview as well?

I live in the US and my wife will be attending interview. Do I need to attend interview with her? Will this effect the interview?

This is posted in the IR1/CR1 section, so I assume your spouse is attending a visa interview at the consulate.  That being said, I am not aware of requirement for the USC spouse to attend.  It can't hurt though.

Yes, but in most countries the US Citizen spouse is not allowed to actually attend the interview, as much due to space limitations as anything else.  It CAN BE helpful to be nearby waiting, if possible, but definitely not technically required.
