Can someone tell me what does my case # ended with 01 means? For example: USA2022123456 01 USA?

Do you have kids who are derivative beneficiaries on your visa case?

How To Read An NVC Case Number (thelawdictionary.org)

The basic NVC case number consists of 13 alphanumeric characters. The first three places are reserved for alphas (letters) identifying the overseas embassy or consulate that received the original immigrant visa application. Some visas are issued according to quotas set for each country.

The next 10 digits of the NVC are numbers that have been coded from the standard Julian calendar. The first four numbers are the year for the application (i.e. 1998). The next three numbers represent the date based on the following formula: Date + 500 (i.e. January 1st would be 001 + 500 = 501). The final number identifies the order of that application for that day (i.e. 4th application = 004.)

and this here explains it a little better I think.

NVC Case Numbers for Immigrants

The case number for your immigrant visa is a 13-digit number comprised of four parts. The number displays as EEEYYYYOOOSSS. EEE is related to the U.S. embassy or consulate that will process your visa. For example, if you are arriving from New Delhi, the first three letters would be NWD. YYYY is the year that the case was received by the NVC. The calendar that the NVC runs is from January to December, not the fiscal year of October to September. OOO is the Julian date (ordinal date, plus 500), and SSS is the sequence number for the day.

I understand that. The extra 01 USA is not part of the 13 digits numbers.

Then I would suspect it's meaningless

The extra 01 usually indicates principal beneficiary's case number, in a case where there are also derivative beneficiaries.  But since your wife's case is CR1, which is not allowed derivatives in the first place, that extra 01 is probably just an error on NVC's part.  Unless your wife answered yes to the question in her DS-260 about having a spouse or children who are immigrating to the US with her?  Do you live outside the US and will you be moving to the US at the same time as your wife?

For both my wifes K1 and my stepsons I130 we just did, they always added a 01 at the end of the NVC case number. With the K1 it was expected as her son would have been 02 if he was on the petition, but they did the same thing on his FX2 Immigrant visa. He was 01 at the end, I think the computer just ads the 01 to the principal case automatically, and would add 02 to the next derivative, the 03, etc.

Here on a K1? Need married and a Certificate in hand within a few hours? I'm here to help. Come to Vegas and I'll marry you Vegas style!!   Visa Journey members are always FREE for my services. I know the costs involved in this whole game of immigration, and if I can save you some money I will!

Both K1 and FX2 categories can have derivatives, so adding 01 to the principal's case would not be strange.  But the OP's wife's case is CR1, which is not allowed derivatives at all.  My case was IR1 and I never saw an extra 01 added to my case number.  It's not added automatically for "immediate relative" visa categories.

My 13 digit NVC case number became a 15 digit visa number when the Consulate placed the visa foil in my passport. .. they added 01. When i used that # while paying for the GC fee, the info said to ignore the last 2 digits.
