No.

 

The DS-5540 is a DHS form, not a USCIS form so doesn’t need to be submitted at the time of filing an I-130.

 

With regards to your other question about whether you or your spouse will be required to bring a completed DS-5540 to the visa interview - email the Embassy and ask.

no actually when you said ds-5540

it confused me and when i got the email they did not mention in the email about ds-5540 so it made me confused still i am confused about what is the complete procedure off that if you can explain in detail thanks

Yes, however that doesn't let you do it whenever you want, it generally needs to be done within 30 days.

When I did it I waited until my spouse was actually immigrating because why spend that money on something that can't be used.

I forget which we used the visa or greencard issuance together with the marriage certificate.
