If they didn't collect biometric info at an earlier time (like at the embassy visa interview), then for sure my wife had her biometrics captured at the port of entry into the USA. 

Now that we've filed the AOS package, I had to pay $85 for a biometric fee, and they will capture them again. Why?

And presumably, for the I-751 ROC in a few years, they will want another fee and capture of biometrics again. 

And possibly, it will happen again if she goes for an N-400 naturalization.

Why the need to repeatedly capture this same information?

Because biometrics is not only composed of a photograph and fingerprints. It is also the process of running those fingerprints through databases in several law enforcement organisations, including the FBI, to confirm the person isn't a wanted individual. And it is for this reason that biometrics are only valid for 15 months, and are required to be re-done at multiple points in the process (and sometimes even in the same process, if USCIS take long enough to adjudicate that particular application).

I was at POE with my K-1 fiancée.  I’ve had green card (twice) and N-400 biometrics. 

 

And I’ve had global entry biometrics collected.  

 

Suffice to say that a USCIS biometrics collection is a much different process than a CBP biometrics collection.

 

I suspect she will do this again on removal of conditions and on N-400. And global entry (absolutely get her global entry if she qualifies now or after she gets her green card(.

I also always assumed it was part of the vetting process while making sure that it is actually the same person at each step of the way.

I hadn't even considered that, but it certainly makes sense. Although it would be hard (but not impossible) to fake fingerprints.

That's definitely one of the reasons - and also why your print shows up on the green card itself. It's a lot more verifiable than a printed signature.
