The question on the D160 form is "Are there other persons traveling with you?"

 

My father is traveling alone as a non-US citizen. But boarding the plane, he will be traveling with a relative (who holds a US passport). 
 

Will the answer be a yes or a no?

The answer will be no. They just want to know if there are other people immigrating together with him. Like a child of his etc.

Travel companion generally means 'family/colleague/tour grp, etc'.  A non-immediate relative should be a 'no' for the ds160 IMO.
