I have mailed off my I-129 application and I was curious to know at what point does the check for the filing fee of $535 gets processed? It would be nice to know for two reasons...1: that my application is not growing mold in a pile somewhere at USCIS and 2: Making sure I keep that extra $535 in my checking account

How long ago did you mail it? Did you send it regular mail, priority, etc.?  If you have just mailed it, it's possible it hasn't reached them yet...normally takes a few days from when they receive it to cash the check....but depending upon your bank, it may take up to at least 5 business days for it to clear.

 

If you purchased tracking from the post office, you can see if they have received it at least and maybe get a better idea of how long you might have to wait...

I mailed it on Friday the 25th and tracking showed that it was received yesterday the 29th.....quicker than I thought so I was happy about that..... I was not sure that they actually wait after the application goes through some kind of review then the check is cashed? or if the check gets cashed at the point when there is actually a case number?

The check gets cashed after they receive your application as properly filed (someone makes sure all the info is filled in, and the application signed).  They send you a receipt notice and begin to process it after that.
