You do not need to contact NVC. You can, if you are impatient and want a number to track you petition enroute from NVC to the embassy. But you do not have to.

General advice is to fill the DS-160 once the case is at the embassy and they contact you - the majority of embassies does that by mailing a letter.

So are there 2 letters after the NOA 2 stage - one going to the petitioner and then one to the beneficiary when it gets to the embassy?

I called NVC because I wanted the case number right away so I could start on the DS-160 when the petition did arrive at the embassy. I would not work on DS-160 though until the embassy as received the petition and ceac.state.gov states it is ready for interview. But having the case number helps a lot. I received the Case Number letter about two weeks after getting case number and also made a request to have it emailed to me, which took 45 days to get back. It is nice to know that your package left NVC and is in route to embassy and when the embassy receives it. But it takes multiple redials to get through...

OK good to know. So you need the case number technically to start the DS-160? They require the number on the form I am assuming?

No, the DS-160 is filled with the SRC number. The case number is required for the medical though.

In Ukraine if you have case number you can go and do medical even before scheduling the interview. You should check your country for specific information. The case number is not needed for ds-160. The petition number given by USCIS is needed.

Could have sworn it asked for case number. But guess I am wrong on that. EDIT: Looking at the print off I made, it does say receipt or petition number. There I used my case number. So you are are right src number is okay :).
