The fingerprints are now digital so if you are on one of the lists they check them against they will know almost immediately. What specific lists they use and whether you are on that list or not is information protected by national security regulations.

Strange question, unless the person involved with the fingerprints is planning on lying about something like a prior visa refusal. Regardless of what lists the fingerprints are compared to, lying during any part of the process is not a good idea. As the visa function is a joint responsibility of the Department of State and USCIS, I think it's a guarantee that USCIS at the port of entry will be compared to the visa applicant databases.
