So once I complete the DS1-160 and schedule

Medical and obtain all other relevant evidence then I should submit the readiness form found at :

Thereafter should I simply call or email the embassy asking for a deferral of interview until

I return in June? On the above form I didn't see any blank spaces to input a request.

Include a Cover Letter with the form.

Write in this Cover letter something along the lines of 'Please find enclosed form DS1-160 indicating my readiness for interview. I completed my medical on xxx date. I am travelling out of the country to visit my fiancee for 2 weeks between x & y dates so I would like to request that my interview be scheduled not before x date and as soon as possible after y date. We are planning to get married in the US on z date in August, 2014. Thank you very much, etc.'

Should I then send a letter separately to the embassy requesting the date for my interview?

OK sorry they've changed the process shortly after I did it (Aug-Sep 2013) and the DS-160 is now submitted online.

I'd send a follow-up letter in writing, or (perhaps better) use the Contact Form on the Embassy website:

http://london.usembassy.gov/niv/visa_contact_form.html
