One petition for the sister. She can not file for her nephew. The petition for her sister will take 12 years. The son will initially be included but he will age out.

Once sister gets her green card, she can file for an unmarried son over 21. That will take 7 years. So, the quickest the nephew can get her through your wife and his mother is 19 years. There is no quicker way through these family connections.
