My wife's uncle in the US has just agreed to be our Joint Sponsor. He's a doctor and has plenty of financial means to meet the minimum requirements.

However, he realized that some years ago he also sponsored another family friend on a work visa (as he remembers it). Are joint sponsors limited in the number of people they can sign-on to sponsor? Or is it only limited to their income and household size?

Nope, he just has to make sure he includes that person in his household size. Is he still in contact with the person? If they abandoned their green card and returned to their home country, or obtained US citizenship, he will not have to count them.

unless that person became a USC, if he is not and continues to be a LPR then he has to count him

the limit is the amount of income, he can be a joint sponsor as many as he wants to, as long as he makes enough income to cover everybody.
