I'm a naturalized U.S. citizen. After I filed I-130 for my brother who resides in his home country ,

he got married. can I upgrade and add his wife to application?

or i must be filed another I-130 because the old form will be Canceled

I filed I-130 in May 2011 and he got married May 2012

please I need advice

no, no need to file another i130 just upgrade the one he has. he still has a very long way to go, so there might be children in the future, you might want to upgrade now, or upgrade later. he still has 10+ years of waiting

You do not need to "upgrade" anything since there is only one category when a USC petitions for a brother.

You can simply write to the government agency where the case is currently (either USCIS or the NVC), provide a PHOTOCOPY of your brother's marriage certificate, a PHOTOCOPY of his wife's birth certificate, English translations of both documents, and a simple paragraph asking she be added to your brother's case.
