They would apply for a visitors visa at their embassy. Nothing you file at all.

It is up to them to file all of the paperwork and provide solid proof of ties to their country that will show their intent to return home before the visa is expired. You don't need to do anything.

But does my Mom need to fill out a Sponsor Form for her Brother and his wife?

Why does your Mom have to fill out anything her brother and his wife?

No your Mom does not have to fill out anything to sponsor her brother and his wife, her brother and his wife would have to apply at consulate in India.

They need to fill out DS-160 and pay the visa fee, schedule an interview and provide documentation like their financial records, investments, holdings etc and any other reason like job, family, kids etc. why they would return back home and all the reason why they would not overstay their visa and return back when their stay is over.
