Hi i was wondering if i can write on the top of my g325a form the word petitioner in pen. So that they can see who is the petitioner and who is the beneficiary more easily. Can i write it in pen??

No need for this at all. They know who is who because of how you fill the forms out. Obviously the US citizen is petitioning for the beneficiary from a foreign country.
