you can pay the visa application fee without the letter. it takes 3 to 4 days for the petition to be sent from NVC to the Embassy. once at the Embassy it take take up to a week to be entered into their system. after that, you can schedule the interview.

she can pay the visa fee even if she has not received the letter yet.Just let her bring her passport and a copy of the biopage of it.I suggest not to wait for letters coz it takes 2 to 3 weeks to get delivered.
